Kodiak Sciences (KOD) is soaring today after announcing that Zenkuda and tabirafusp-ted each met their primary endpoints in the Phase 3 DAYBREAK study in wet age-related macular degeneration (wAMD). KOD is a precommercial, retina-focused biotech developing three late-stage programs, with lead candidate Zenkuda targeting wAMD, diabetic retinopathy and retinal vein occlusion. The readout moves KOD another step closer to potentially commercializing its first product, with Zenkuda now supported by five positive Phase 3 studies ahead of a planned 4Q26 BLA filing.
- Zenkuda: Demonstrated meaningful vision gains alongside rapid and sustained retinal drying, with 54% of patients reaching 6-month dosing under strict treat-to-dryness retreatment criteria. DAYBREAK adds to positive Phase 3 results from DAYLIGHT in wAMD, GLOW and GLOW2 in diabetic retinopathy and BEACON in retinal vein occlusion.
- Tabirafusp-ted: Also met its vision primary and anatomical key secondary endpoints. The bispecific targets VEGF and IL-6, with KOD planning post-hoc analyses to identify wAMD subgroups that may benefit from IL-6 inhibition. It is also advancing in Phase 3 ALTO in diabetic macular edema, designed to test superiority versus aflibercept.
- Market opportunity: KOD says Zenkuda and tabirafusp-ted target the roughly $15 bln anti-VEGF market across retinal vascular diseases. DAYBREAK was designed to strengthen Zenkuda's competitive position in wAMD.
- Next steps: KOD plans to submit a three-indication Zenkuda BLA in 4Q26 supported by five positive Phase 3 studies. Beyond Zenkuda, Phase 3 PEAK data for KSI-101 in macular edema secondary to inflammation are expected in December, with a KSI-101 BLA planned for 2Q27. KOD believes Zenkuda and KSI-101 could potentially receive marketing authorization by the end of 2027 or early 2028.

KOD is seeing an outsized move today as the DAYBREAK results move the company another step closer to potentially commercializing its first product. Zenkuda is KOD's lead candidate and is now supported by five positive Phase 3 studies ahead of a planned 4Q26 BLA, with its durability profile in wAMD emerging as an important potential differentiator. Specifically, 54% of DAYBREAK patients reached a 6-month dosing interval while maintaining vision gains and retinal drying under strict retreatment criteria, highlighting the potential to reduce treatment burden without sacrificing disease control. The readout increasingly shifts the Zenkuda story away from proving clinical efficacy and toward regulatory approval, the breadth of an eventual label and KOD's ability to execute commercially. DAYBREAK also strengthens the broader pipeline story, with tabirafusp-ted meeting its primary endpoint and providing additional validation for KOD's late-stage retinal platform. The candidate is now advancing in Phase 3 ALTO in diabetic macular edema, where KOD is testing whether its dual VEGF/IL-6 approach can deliver superior outcomes versus aflibercept. Meanwhile, KSI-101 provides another late-stage opportunity, with Phase 3 PEAK data expected in December.
